Obviously that's some kind of rhyming slang for lovers' lane that he's picked up somewhere in backwoods America during his travels. And he's literally in a supernaturally strong man's arms -- not Superman's, but a superman's. Castiel's, actually. You see? It's all very simple if you just read it right. *nods wisely*

However, I admit I'm not sure what the "manly hood" is. I'm leaning towards Dean's foreskin at this point, but that rather begs the questions of where it's been that he wants it back...? Other than his own throat, which is kind of what the sentence implies.
